Abstract Objectives The aim of this study was to examine the changes in gray matter in nasopharyngeal carcinoma patients with normal hearing (Group 1) and nasopharyngeal carcinoma patients with hearing loss (Group 2) after radiotherapy using voxel-based morphological analysis and to analyze the relationship with the radiation doses of the temporal lobe. Methods 21 patients in Group 1, 14 patients in Group 2, and 21 healthy volunteers were selected. All participants underwent an otologic examination and three-dimensional magnetization preparatory rapid acquisition gradient echo sequence scan. The correlation between the variation of whole brain gray matter volume and the doses of the temporal lobe was analyzed by Data Processing & Analysis for Brain Imaging software. Results Compared with the normal control group, the brain areas with reduced gray matter volume in nasopharyngeal carcinoma patients after radiotherapy were mainly in the left posterior cerebellar lobe (T = −8.797), left insular lobe (T = −7.96), and the right insular lobe (T = −6.632). Compared to Group 1, the brain areas of Group 2 patients with reduced gray matter volume were mainly in the left superior temporal gyrus (T = −2.366), left olfactory bulb (T = −2.52), left Rolandic operculum (T = −2.431), and right olfactory bulb (T = −3.100). Compared with Group 1, the brain areas of Group 2 patients with increased gray matter volume were mainly in the left calcarine sulcus (T = 3.425) and right calcarine sulcus (T = 3.169). There were no correlations between the changes of brain gray matter volume and the radiation doses of the temporal lobe in both Group 1 and Group 2. Conclusions The radiotherapy may cause the changes of brain areas associated with cognitive function in nasopharyngeal carcinoma in a long-term follow-up. At the same time, nasopharyngeal carcinoma patients with the radiation-induced hearing loss had abnormal gray matter volumes in the auditory center and other sensory centers. Our findings might provide new understanding into the pathogenesis of radiation-induced brain damage in normal-appearing brain tissue. Yet this exploratory study should be taken with caution.

Objective:To explore an assay that can concisely, rapidly, and accurately quantify the amount of chimeric antigen receptor (CAR)-T cells in the bone marrow or peripheral blood of patients after CAR-T cell immunotherapy by morphological analysis and flow cytometry assay, providing timely and accurate feedback for clinical treatment.Methods:We analyzed the CAR-T cell detection results in peripheral blood and bone marrow of 256 patients who received CAR-T cell immunotherapy in the Department of Hematology, Affiliated Hospital of Xuzhou Medical University from August 2016 to August 2021. All 256 patients survived more than one month after CAR-T cell infusion. Among them, there were 118 patients with multiple myeloma, 68 patients with acute lymphoblastic leukemia, and 70 patients with lymphoma. The morphological characteristics, positive rate and detection rate of CAR-T cell in peripheral blood and bone marrow were analyzed by morphological methods. The positive rate and detection rate of CAR-T in peripheral blood and bone marrow were analyzed by flow cytometry protein L detection. χ 2 test was used to comprehensively analyze the difference between the detection rate of the combined analysis of the two methods and the detection rate of the single method. Results:CAR-T cells have significant morphological characteristics, and there are obvious morphological differences from normal lymphocytes. The detection rates of CAR-T cells in peripheral blood or bone marrow by morphological methods and flow cytometry were 88.28%(226/256) and 79.29% (203/256), respectively. When the two methods were combined, the detection rate of CAR-T cells can reach 99.22%, with statistically significant difference comparing to that of single method( P<0.05). Through the analysis of the detection results of peripheral blood at different time points, it was found that the average detection rates of morphology and flow cytometry in 118 patients with multiple myeloma were 9.50% and 10.23% on the 7th day, and 13.50% and 15.19% respectively on the 15th day. On the 21st day, the average detection rates of morphology and flow cytometry were 8.00% and 10.07%, respectively. The average detection rates of morphology and flow cytometry in 68 patients with acute lymphoblastic leukemia were 12.00% and 11.22% on the 7th day, and 21.00% and 23.10% respectively on the 15th day. On the 21st day, the average detection rates of morphology and flow cytometry were 13.50% and 10.91%, respectively. The average detection rates of morphology and flow cytometry in 70 lymphoma patients were 7.50% and 10.35% on the 7th day, and 9.00% and 10.35% respectively on the 15th day. The average detection rates of morphology and flow cytometry at 21 days were 6.50% and 5.69%, respectively. The number of CAR-T cells in samples from patients with different diseases reached a peak around the 15th day. Conclusion:The detection rate of CAR-T cells from peripheral blood or bone marrow was significantly higher with the combination of the 2 methods compared to the single method.

Introducción: La anemia drepanocítica es una anomalía genética hereditaria de la hemoglobina, que se caracteriza por la presencia de glóbulos rojos que pierden su forma redonda característica y adquieren forma de hoz. Aunque aún no tiene cura definitiva, se desarrollan varias acciones con el propósito de mejorar la calidad de vida y la atención médica a los pacientes. Objetivos: Conocer los aspectos referidos al análisis automatizado de formas en eritrocitos en los últimos años y proporcionar una visión en el caso de la drepanocitosis, que permita determinar las limitaciones actuales, principalmente para el empleo de herramientas automatizadas en el seguimiento clínico de pacientes con esta enfermedad. Método: Se realizó la revisión sistemática de la literatura de los años 2018, 2019 y dos aportes del 2020, en tres bases de datos electrónicas de amplio alcance: IEEEXplore, Google Scholar y SCOPUS. Los documentos se analizaron teniendo en cuenta preguntas específicas para obtener criterios generales sobre la situación de interés. Conclusiones: Los análisis realizados revelan un volumen creciente de investigaciones en este campo, con resultados de varios países. El examen detallado de las investigaciones permitió identificar problemas referidos a las métricas de evaluación empleadas, a los algoritmos para el análisis y procesamiento de imágenes, empleo del criterio médico, bases de datos empleadas y, herramientas para el análisis automático de formas de eritrocitos(AU)


Introduction: Sickle-cell anemia is a genetic hereditary anomaly of hemoglobin characterized by red blood cells that lose their normal round morphology and acquire a sickle shape. Although no cure is so far available, several actions are in progress to improve the quality of life and medical care of patients. Objective: Become acquainted with aspects related to the automated morphological analysis of erythrocytes in recent years, particularly in the context of sickle-cell anemia, allowing to determine the current limitations, mainly in the use of automated tools for the clinical follow-up of sickle-cell anemia patients. Methods: A systematic review was conducted of the literature published in the years 2018, 2019, and two contributions from 2020, in three broad scope electronic databases: IEEEXplore, Google Scholar and SCOPUS. The documents were analyzed on the basis of specific questions to obtain general criteria about the situation of interest. Conclusions: The analysis conducted revealed a growing volume of research in this field, with results in several countries. Detailed examination of the studies led to identification of problems related to the evaluation metrics used, the algorithms for the analysis and processing of images, the use of medical criteria, the databases used and tools for the automated morphological analysis of erythrocytes(AU)

ABSTRACT

The penis represents the organ of the male's copulation. It is essential to know the reproductive biology and the morphology of the reproductive organs to increase animal production. In order to contribute to this knowledge and provides information on the ram reproductive morphology, the purpose of this work was to describe the distribution, based on light microscopy, of the collagen and elastic fibers in the ram penis. For that, were collected transverse fragments of the penis (root, sigmoid flexure, body and glans) of seven rams. The specimens were fixed in paraformaldehyde for 24h and destined for the histological routine. The extracellular matrix of the ram penis was composed of collagen and elastic fibers. The penis was enveloped by the tunica albuginea, consisting essentially of collagen fibers, which were arranged in two layers: an outer longitudinal and an inner circular. This tunic emitted septa that penetrated the corpus cavernosum. The elastic fibers appeared transversely and longitudinally in the corpus cavernosum, corpus spongiosum, and next to the neurovascular bundle of the penis. This structure was not different to that reported for other domestic ruminants such as cattle and buffaloes.(AU)


O pênis representa o órgão da cópula do macho. É fundamental que se conheça a biologia reprodutiva e a morfologia dos órgãos reprodutores para o incremento da produção animal. Com o objetivo de contribuir para este conhecimento e fornecer informações sobre a morfologia reprodutiva de ovinos, o propósito deste trabalho foi descrever a distribuição, com base na microscopia de luz, das fibras colágenas e elásticas no pênis de ovinos. Para tanto, foram coletados fragmentos transversais do pênis (raiz, flexura sigmoide, corpo e glande) de sete ovinos. Os exemplares foram fixados em paraformaldeído por 24h e destinados à rotina histológica. A matriz extracelular do pênis de ovinos estava constituída por fibras colágenas e elásticas. O pênis estava envolvido pela túnica albugínea, formada essencialmente por fibras colágenas, que estavam arranjadas em duas camadas: longitudinal externa e circular interna. Esta túnica emitiu septos que penetraram no corpo cavernoso. As fibras elásticas apareceram de modo transversal e longitudinal nos corpos cavernoso e esponjoso e junto ao feixe vásculo-nervoso do pênis de ovinos. Esta estrutura mostrou-se similar ao encontrado em outros ruminantes domésticos como bovinos e bubalinos.(AU)

ABSTRACT

Se realizó un estudio descriptivo y transversal desde julio hasta octubre de 2017, por especialistas de la Universidad de Oriente y de la Universidad de Sao Paulo, Brasil, para analizar desde el punto de vista morfológico células endoteliales de venas del cordón umbilical humano, presentes en imágenes digitales de cultivos in vitro 2D, tratadas con la ß2GPI. . Se propuso la clasificación supervisada celular considerando 3 clases: circulares, deformadas alargadas y deformadas poco alargadas, según los coeficientes de formas elíptico y circular, todo lo cual permitió identificar formas celulares relevantes. Para comparar los resultados de las muestras de control y las tratadas, se calcularon los intervalos de confianza para cada una de las clases, con un nivel de confianza de 95 por ciento. Se concluye que el análisis de las alteraciones morfológicas in vitro puede ser utilizada en cultivos 2D precoces (de 24 y 48 horas) para la cuantificación de la angiogénesis


A descriptive and cross-sectional study was carried out from July to October, 2017, by specialists of the Oriente University and the University of Sao Paulo, Brazil, to analyze from the morphological point of view endothelial cells of the human umbilical cord veins, which were present in digital images of 2D in vitro cultures, treated with the ß2GPI. The cellular supervised classification was proposed considering 3 classes: circular, distorted elongated and distorted not very elongated, according to the coefficients of elliptic and circular shapes, all that allowed to identify outstanding cellular forms. To compare the results of the control and treated samples, the intervals of confidence were calculated for each of the classes, with a 95 percent level of confidence. It was concluded that the analysis of the morphological disorders in vitro can be used in early 2D cultures (24 and 48 hours) for the quantification of the angiogenesis

Introduction/Aim: Aging of the parahippocampal cortex is accompanied by atrophy. The most common changes include cell number reduction, formation of corpora amylacea in astrocytes, which may represent the basis for the development of cognitive dysfunction. The aim of this study was to detect presence and distribution of corpora amylacea in subpial and subcortical region of parahippocampal cortex during aging process using histochemical analysis as well as to carry out quantification of structural changes in different cortical layers using histochemical and immunohistochemical methods. Material and Methods: Material represents tissue of uncus of parahippocampal gyrus of right cerebral hemisphere of 30 cadavers taken from autopsies carried out at the Forensic Medicine Institute of the Niš Faculty of Medicine. Upon tissue preparation using standard histological procedures, 10 μm thick sections were made and stained with hematoxylin eosin and periodically with acid Schiff's method. The total number of 30 samples, 16 to 93 years of age, was analyzed. Results: During the normal aging process in parahippocampal cortex the number of corpora amylacea localized in subpial region increases, and it is even greater in subcortical white matter, with no significant change in size and shape. Positive reaction to neuron specific enolase, determined by immunohistochemical methods suggests the presence of neural components, while a weaker reaction to glial acid protein and positive one to S100 protein suggests the possibility that the corpora amylacea are astrocyte inclusion. Conclusion: The obtained results indicates that the presence of these structural changes may lead to damage of those brain structures responsible for normal memory function and thus responsible for cognitive changes present in healthy elderly individuals.

ABSTRACT

La formación de cálculos en el sistema urinario es característica de la urolitiasis. Cuando los cálculos son mayores de 20 mm de largo se consideran voluminosos y coraliformes cuando llenan la pelvis renal y se ramifican en uno o más cálices. Los cálculos urinarios frecuentemente pueden comprometer la integridad y funcionalidad renal. Este estudio descriptivo y retrospectivo tiene como objetivo describir la composición de cálculos voluminosos y coraliformes obtenidos de 67 pacientes que concurrieron al IICS de marzo del 2007 a abril del 2011 y cumplieron con los criterios de inclusión. El análisis morfológico de los cálculos se realizó aplicando criterios mineralógicos y morfológicos descritos por Daudon et al. dando diferentes tipos morfológicos, cada uno con una composición dominante y una causa etiológica probable para la formación del cálculo. Los tipos más frecuentes en el núcleo fueron IVa1 y IIa compuestos respectivamente de carbapatita (fosfato de calcio) y Weddellita (oxalato de calcio dihidratado), mientras que Ia compuesto por Whewhellita (oxalato de calcio monohidratado) fue el tipo más frecuente en sección y superficie. En el 87% de los cálculos se detectó la presencia de 2 o más tipos morfológicos asociados, mostrando la etiología multifactorial de la urolitiasis. La cirugía abierta fue la técnica más frecuentemente empleada para remover el cálculo, 5 pacientes perdieron un riñón como consecuencia de la litiasis. Estos resultados apuntan a la importancia de la identificación correcta de la causa que genera los cálculos para seleccionar medidas apropiadas para evitar la recurrencia y las complicaciones severas.


The formation of stones in the urinary system is characteristic of urolithiasis. When thestones are bigger than 20 mm long they are considered voluminous and staghorn whenthey fill the renal pelvis and branch into one or more of the calices. Urinary stones could frequently compromise renal integrity and functionality. This descriptive and retrospective study aimed to describe the composition of voluminous and staghorn stones obtained from 67 patients who attended IICS from March, 2007 to April, 2011 and complied with the inclusion criteria. Morphological analysis of kidney stones was made by applying mineralogical and morphological criteria described previously by Daudon et al. giving different morphological types, each one with a dominant composition and a probable etiological cause of the stone formation. The more frequent types in the nucleus were IVa1 and IIa composed respectively of carbapatite (calcium phosphate) and weddellite (calciumoxalate dihidrated) while Ia composed of Whewhellite (calcium oxalate monohydrated) wasthe more frequent type in section and surface. The presence of 2 or more morphological types associated was detected in 87% of the stones, showing the multifactorial etiology of urolithiasis. Open surgery was the more frequent technique to remove the stones 5 patientslost one kidney as an outcome of lithiasis. This results point the importance of the correct identification of the cause that generates the stones to select proper measures to avoid recurrence and severe complications.

ABSTRACT

Desde tiempos antiguos la medicina tradicional ha usado extensamente las especies del género Lippia como analgésicos, antiinflamatorios, antipiréticos, antifúngicos, etc. Numerosos estudios describen diversos compuestos presentes en extractos acuosos, metanólicos, o aceites esenciales de estas plantas, siendo los flavonoides los compuestos más abundantes. Sin embargo, la composición y cantidad de los metabolitos secundarios dependen de la zona geográfica, factores climáticos, altitud, época de cosecha y estado de crecimiento de estas plantas. El objetivo de este estudio fue evaluar la actividad antioxidante del extracto acuoso de orégano (Lippia graveolens HBK) del Norte de México y su efecto tóxico in vitro e in vivo. La capacidad antioxidante del extracto acuoso se midió por el método de DPPH en seis diluciones del extracto (5-160 mg/mL) y se utilizó Trolox como referencia; para el efecto tóxico in vitro se usó el ensayo de citotoxicidad con larvas de Artemia salina. Para el efecto in vivo se emplearon 24 ratones árabes machos divididos en 6 grupos de animales (n=4): 4 grupos experimentales con 10, 100, 1000 y 5000 mg del extracto/ kg de peso administrados vía oral respectivamente, además de un grupo control positivo (5 mg de colchicina/kg de peso vía i.p) y un grupo control negativo (solo agua destilada). Después del tratamiento los ratones se sacrificaron y se colectaron muestras de hígado y riñón que se sometieron a estudios histológicos e histoquímicos, además se realizó un análisis cuantitativo. La actividad antioxidante del extracto acuoso de orégano se presentó a 160 mg/mL. La CL50 fue mayor a 1,000 ug/mL por lo que el extracto se considera no tóxico. En el análisis morfológico in vivo con distintas dosis del extracto acuoso de orégano no se observó un efecto tóxico. Since ancient times, traditional medicine has widely used species of the genus Lippia as analgesics, anti-inflammatory, antipyretic, antifungal, etc. Numerous studies describe several compounds present in aqueous extracts, methanol, or essential oils of these plants, being flavonoids the most abundant compounds. However, the composition and quantity of secondary metabolites depend on the geographical area, climatic factors, altitude, time of harvest and growth status of these plants. The objective of this study was to evaluate the antioxidant activity of aqueous extract of oregano (Lippia graveolens HBK) from the North of Mexico and its toxic effect in vitro and in vivo. The antioxidant activity of the aqueous extract was measured by DPPH method in six dilutions of the extract (5-160 mg / mL), Trolox was used as a reference. For the in vitro toxic effect, cytotoxicity assay with larvae of Artemia salina was used. For the in vivo effect, 24 males mice were used and divided into 6 groups (n = 4): 4 experimental groups with 10, 100, 1000 and 5000 mg extract / kg body weight administered orally respectively, also we used a group positive control (5 mg of colchicine / kg body weight administered via ip) and a negative control group (distilled water only). After treatment all mice were sacrificed, and samples from liver and kidney were collected and analyzed by histological and histochemical studies. Also a quantitative analysis was done. The antioxidant activity of aqueous extract of oregano was presented at 160 mg/mL. The LC50 was greater than 1.000 mg/mL, so the extract is considered nontoxic. In the morphological analysis in vivo with different doses of aqueous extract of oregano, no toxic effect was observed. The results validate the use of oregano in traditional medicine...

Determinar criterios de evaluación morfológica del hemograma, basados en parámetros clínicos y resultados del contador hematológico, en términos de gestión, costos y clínica en un hospital tipo 1. Pacientes, materiales y métodos: muestras de hemogramas de pacientes del hospital de carabineros de Chile durante el mes de agosto de 2009, analizadas en un equipo HMX Beckman-Coulter y evaluadas morfológicamente por Tecnólogo Médico(TM). Se registraron parámetros clínicos como hemoglobina, leucocitos y plaquetas; además se consideró el lugar de derivación, especialidad y edad del paciente. Se determinaron los costos e ingresos en función de los resultados obtenidos. Resultados: de los 2.222 pacientes evaluados, existió concordancia entre el Tecnólogo Médico y el analizador en 93,3 por ciento de los casos, con una sensibilidad de 97,3 por ciento y una especificidad de 88,9 por ciento con valores predictivos positivo y negativo de 95,9 por ciento y 92,7 por ciento respectivamente, para detectar una muestra anormal. Con los criterios de selección fue posible eliminar la evaluación morfológca en 79,5 por ciento de los casos, permitiendo un ahorro de $3655929 en el mes por concepto de hemogramas. Discusión: el estudio demostró que es posible disminuir el número de evaluaciones morfológicas realizadas al hemograma a valores similares obtenidos en estudios internacionales, con reducción significativa de los costos y tiempos de TM.


Objective: to determine criteria for the morphological analysis of the full blood count, based on the clinical parameters and the results of the full blood count using an automatic counter in terms of cost and clinicla implications in a distric hospital. Patients, Materials and Method: Full blood count samples from patients attended in the Hospital de Carabineros de Chile during the month of August 2009 were analyzed using HmX Beckman-Coulter and evaluated morpholigically by the Medical Science Officer(Medical Technologist). Clinical parameters, hemoglobin, white cell count platete count were registered, as well the origin of the specimen, medical speciality an patient age. The cost an income derived from the full blood count were determined by the Unit of Cost, Hospital of Carabineros and the changes in costs calculated in function of the results obtained. Results: Of the 2,222 patients evaluated, there was a concordance between the Clinical Science Officer(Medical Technologist) and the automatic counter in 93,9 percent of the samples, with a sensitivity of 97,3 percent and a specificy of 88,9 percent, a positive and negative predictive evalues of 95,9 percent and 92,7 percent respectively for detecting an abnormal sample. Using the criteria of selection it was possible to eliminate the evaluation morphological in 79,5 percent of the samples permitting a saving of $3655929 per month for the analysis of full morphological evaluations of the blood count without losing clinically important data in 80 percent of the samples, which is similar to that reported internationally, with a significant decrease in cost an Medical Scientific Officer time.

ABSTRACT

O alvo deste estudo foi apresentar um novo método de avaliação das regiões organizadoras de nucléolos (NORs), objetivando verificar a correlação entre malignidade das células tumorais com proliferação celular. Vinte e nove tumores foram analisados quanto aos aspectos morfológicos e quantitativos de regiões organizadoras de nucléolos coradas pela prata (AgNORs) e comparados à quatro glândulas mamárias normais utilizadas como controle. Os dados obtidos foram tratados pelos testes não-paramétrico de Kruskal-Wallis e ôUõ de Mann-Whitney. Os números médios de NORs dos grupos neoplásicos malignos incluindo carcinomas tubulares (2,14 ± 0,54), carcinomas papilíferos (1,79 ± 0,24) e carcinomas sólidos (1,95 ± 0,30) mostraram diferença significativa quando comparados ao grupo controle (1,16 ± 0,08). A análise morfológica das AgNORs mostrou que o padrão de apresentação da NORs do tipo I prevaleceu nos grupos neoplásicos benignos (75% dos casos de adenoma tubular e 50% dos casos de carcinoma tubular), enquanto o tipo II prevaleceu nos grupos neoplásicos de comportamento maligno (54,5 % dos carcinomas papilíferos e 100% dos carcinomas sólidos). O grupo controle mostrou na sua totalidade (100% dos casos) predominância de NORs do tipo IV. O padrão do Tipo III não predominou em nenhum dos grupos envolvidos neste estudo. A análise da morfologia das AgNORs é uma ferramenta útil na determinação do prognóstica de alterações neoplásicas da glândula mamária canina.


The aim of this study was try to present a new method of evaluation of the nucleolar organizer regions (NORs) being related its morphology and distribution, objectifying to verify the correlation between malignancy of tumoral cells with cellular proliferation. Twenty-nine tumors were analyzed with regard to quantitative and morphological aspects of the silver staining nucleolar organizer regions (AgNORs) and compared to four normal mammary glands used as control. The data obtained were submitted to non-parametric Kruskal- Wallis and Mann-Whitney ôUõ tests. The mean NORs counts of the malignant neoplastic groups tubular carcinomas (2.14 ± 0.54), papillary carcinomas (1.79 ± 0.24) and solid carcinomas (1.95 ± 0.30) showed significant difference when compared with the control group (1.16 ± 0.08). Morphological analysis of AgNORs revealed that the expression pattern of type I NORs predominate in the benign histological groups (75% of the tubular adenoma cases and 50% of the tubular carcinoma cases), whereas type II NORs pattern prevailed in the malignant behavior neoplastic groups (54,5% of the papillary carcinoma and 100% of the solid carcinoma cases). Control group showed total predominance (100% of the cases) of type IV NORs. Expression pattern of the type III NORs did not prevail in neither histological group encompassed by this study. Evaluation of AgNORs morphology reveals an useful tool for prognosis assessment of neoplastic alterations of the canine mammary gland.

ABSTRACT

The morphology and location of the pterion was investigated in total of 149 Korean skulls (298 pteria). The most common form of the pterion was a sphenoparietal articulation in the incidence of 76.5%. The most common location of the pterion was in an area which covers 5 mm from the supraorbital horizontal line parallel to Frankfurt line and the incidence was 81.8%. The metric location of the pterion was on 36.9 +/-3.8 mm above the upper margin of the zygomatic arch and 26.8 +/-4.5 mm behind the posterior margin of the frontozygomatic suture. The average length of the pterion was 12.2 +/-4.4 mm. The most common form of the epipteric bone was a single bone which articulated with the frontal, parietal, temporal and sphenoid bone, and the incidence was 40.3%. The most common location of the epipteric bone was in an area which covers 5 mm from the supraorbital horizontal line parallel to Frankfurt line and the incidence was 45.2%. Morphological analysis of the pterion, when combined with some other non -metric and metric variants, may become useful physical anthropologic tools for the identification of the Korean skull.
